Brightseed, a company specializing in bioactive compound discovery using artificial intelligence, has provided preclinical data showing that two compounds found in hemp hulls, N-trans caffeoyltyramine (NCT) and N-trans feruloyltyramine (NFT), have potential to support gut barrier function.
The gut barrier plays a role in maintaining overall digestive and immune function, as it enables nutrient absorption and acts as a barrier against pathogens and toxins. Increased permeability of the intestinal epithelium, commonly referred to as “leaky gut,” may be associated with impaired gut health.
In the in vitro study, NCT and NFT were found to improve impaired gut barrier function in a culture of human primary intestinal epithelial cells, which was produced using RepliGut, a validated model for intestinal epithelium.
Administering both of the bioactives to the inflamed cells demonstrated a statistically significant reversal of impaired intestinal permeability and transepithelial electrical resistance (TEER).

Brightseed’s AI platform, Forager, discovered NCT and NFT’s activity on a biological receptor governing gut health. The two compounds are agonists for the HNF4a protein. Decreased expression of HNF4a in the gut is linked to a high-fat diet, poor intestinal health, and decreased gut barrier integrity.
In the fall of 2022, Brightseed launched Bio 01, an upcycled hemp-based dietary fiber ingredient containing NCT and NFT, as a gut support ingredient designed to fortify cereals, granolas, nutrition bars, functional beverages, and other applications.
